Seems you are missing value for mandatory field. Have you customized Production Order form?

I am unable to Submit the production order & without submitting I cannot post the manufacturing entry based on BOM from Material Transfer Option in Stock.

I could not reproduce the issue. Have you customized Production Order? Is there any workflow on PO?
